Nonstop mutation in the kisspeptin 1 receptor (kiss1r) gene causes normosmic congenital hypogonadotropic hypogonadism

Purpose: Congenital hypogonadotropic hypogonadism (CHH) is a rare genetic disorder mostly characterized by gonadotropins release and/or action deficiencies. Both isolated (idiopathic hypogonadotropic hypogonadism) and syndromic (Kallmann) forms are identified depending on the olfactory ability. Clinical and genetic heterogeneities of CHH have been widely explored, thus improving our understanding of the disease's pathophysiology. This work aims to (1) provide a detailed clinical and hormonal description of normosmic CHH patients and (2) identify the mutation linked to the studied phenotype. Participants and methods: We investigated three affected patients with normosmic CHH, belonging to a consanguineous Tunisian family. Patients underwent an insulin-induced hypoglycemia test. We performed whole exome sequencing to identify the causal mutation. Results: At first diagnosis, a total gonadotropic deficiency was identified in all patients. The insulin-induced hypoglycemia test has also revealed a reduced cortisol secretion and complete growth hormone deficiency. At 20.8 years, one female exhibited a spontaneous recovery of the hypothalamic-pituitary-adrenal axis function, unlike her affected siblings who still depend on corticosteroid replacement therapy. Herein, we identified a novel homozygous nonstop mutation (c.1195T>C) in KISS1R gene in all affected subjects. This mutation led to the substitution of the physiologic stop codon by an arginine (p.X399R). Conclusions: Our study highlights the importance of the KISS1R signaling, in gonadotropin-releasing hormone neurons, in the control of reproductive function. Additionally, our data suggests a complex central and peripheral metabolic control of puberty, through the hypothalamic KISS1R signaling. We suggest a mutual link between the hypothalamic-pituitary-gonadal, -adrenal, and -somatotropic axes.

Phytochemical and therapeutic effect of araucaria columnaris resin extracts on clinical pathogens

Araucaria columnaris is one of the ornamental plants of all over the world. It is commonly known as Christmas tree. In the present study tree exudates – resin from the bark of the Araucaria columnaris belonging to the family Araucariaceae were investigated for phytochemical and antibacterial activity. The resin was separately extracted with different solvents based on its polarity, water, methanol, ethyl acetate and benzene. The extracts were screened for phyto-chemical analysis, the result shows that methanolic extract involved in extraction of large number of phytochemicals when compared with other extracts and subjected to antibacterial assay against major human pathogens. Methanolic extract act as good inhibitory agent against major microorganisms. This finding revealed that methanolic resin extract of Araucaria columnaris had significant potential for the control of dreadful diseases causing human pathogens.

Critical process parameters evaluation of modified nanoprecipitation method on lomustine nanoparticles and cytostatic activity study on l132 human cancer cell line

This work was focused on identification and evaluation of process parameters of modified nanoprecipitation method, for fabrication of lomustine nanoparticles, with the aim of reducing cancer cell viability at low concentration of lomustine. The parameters controlling particle size, mostly in nanosize, were solvent/nonsolvent composition and emulsification speed of homogenizer along with aqueous phase volume. This controlled particle size is below 250 nm. The stabilizer concentration controlled particle size is within 68 nm ± 0.89 to 137 ± 0.94 nm with PDI 0.06 ± 0.008 to 0.25 ± 0.001. But, the stabilizer addition mode showed more uniform size distribution with PDI 0.085 ± 0.004. Entrapment efficiency was maintained well above 47 ± 0.23%. The drug release pattern was monophasic with controlled release over 24 hrs. In the method used, drug content was affected by ratio of polymer to drug to organic solvent, as well as homogenization speed and time. Percentage viable cells of L132 human lung cancer cell line remained, were only 5% at 100 μg/ml lomustine equivalent PLA nanoparticles.

Notre entreprise, dans le présent article, vise à rendre compte de cette spécificité discursive en interrogeant trois expressions ponctuant souvent son discours. Notre analyse se propose d’examiner des segments de discours collationnés à partir des différents entretiens périodiques accordés aux médias nationaux et internationaux ainsi que des allocutions du président lors des travaux réunissant le gouvernement et les walis. En guise de conclusion, nous déterminerons les motivations du choix de ces formules dans le discours présidentiel.

The objective of this research paper is to assess the mediating effect of Hygiene and motivator factors of Herzberg’s theory in motivating employees with special reference to retail industry and estimate their level of job satisfaction through them. The research being descriptive in nature is based on Primary as well as Secondary data. Structured and standard Questionnaire based on motivatorshygiene has been used for data collection. The respondents (retail executive) respondents were contacted through online Google form and questionnaires were distributed by the researcher personally . The final sample size was 180. Factor analysis is the main statistical tool used to identify underlying variables, or factors, that explain the pattern of correlations within a set of given variables. Researcher has used Principal component method of extraction. The KMO Test was carried out for finding sampling adequacy and Bartlett's Test of Sphericity was carried along with approx. Chi-Square test to test the hypothesis. It is identified from the analysis that out of the eight factors six factors are hygiene factors. Out of these six hygiene factors 3 are money based- Monetary gains attracts, Pay Satisfaction and Love of Money. The results have showed that hygiene factors dominated over motivators in terms of job satisfaction (retail executives in NCR. ) . Herzberg’s two factor theory is applicable but not all factors influence or impact and few are becoming obsolete.

Risk of pulmonary-reproductive dysfunctions, inflammation and oxidative dna damage in exposure to polycyclic aromatic hydrocarbon in cigarette smokers

Background: Exposure to cigarette smoke has been associated with pulmonary and reproductive dysfunctions; inflammatory response, oxidative stress and oxidative DNA damage induced by polycyclic aromatic hydrocarbons (PAHs) present in cigarette smoke have been implicated in the pathogenesis of these disorders. The peak expiratory flow rate (PEFR), a biomarker of inflammation and oxidative DNA damage (8-hydroxy-2-deoxyguanosine (8-OHdG), tumor necrosis factor alpha (TNF-α)), reproductive hormones (testosterone (TST), luteinizing hormone (LH), follicle stimulating hormone (FSH)) cotinine and urinary PAH metabolite (1- hydroxypyrene (1-HOP)) were estimated in male active smokers. Methods: One hundred men aged 20-47 years, comprising 50 active male smokers and 50 non-smokers, were randomly recruited into this comparative cross-sectional study. The PEFR was measured using a peak flow meter, serum levels of cotinine, FSH, LH, TST, TNF-α, and urine 8-OHdG by enzyme-linked immunosorbent assay and 1-HOP by high-performance liquid chromatography. Data analysis was done using a t-test and correlation analysis at p≤0.05. Results: Smokers had significantly higher cotinine (49.73±31.76 versus 0.51±0.69 ng/ml, p≤0.001), 8-OHdG (16.34±12.10 versus 5.79±2.14 ng/ml, p≤0.001) and lower PEFR (309.20±56.05 versus 452.80±45.76 L/min, p≤0.001) and LH (5.75±2.06 versus 6.97±2.79 mIU/ml, p=0.015) compared to non-smokers. Duration of exposure to cigarette smoke correlated positively with cotinine (r=0.937, p≤0.001) and 1-HOP (r=0.813, p≤0.001) while cotinine correlated positively with 1-HOP (r=0.863, p≤0.001) only in smokers. Conclusion: Reduced lung function and luteinizing hormone and concurrent increase in oxidative DNA damage associated with exposure to cigarette smoke may suggest the involvement of PAH-induced DNA damage in the development of pulmonary and reproductive impairment in smokers.

Assessment of the nutritional composition, microbiological and sensory attributes of maize-based fortified food with peanut and carrot blends

This research assessed the nutritional composition, microbiological and sensory attributes of maize-based fortified food with peanut and carrot blends. A total of six composite flour samples were formulated using varying ratios of maize, peanut, and carrot: (MPC1) 100% maize flour (control), (MPC2) 90% maize, 5% peanut, and 5% carrot, (MPC3) 80% maize, 15% peanut, and 5% carrot, (MPC4) 70% maize, 20% peanut, and 10% carrot, (MPC5) 60% maize, 25% peanut, and 15% carrot, and (MPC6) 50% maize, 30% peanut, and 20% carrot. The proximate, mineral, vitamin, antinutrient, and functional properties of these fortified food samples were assessed using established methodologies. The proximate analysis revealed a significant increase (p<0.05) in moisture, crude protein, fat, crude fibre, ash, and energy content of the fortified samples, with values ranging from 1.63% to 3.80%; 3.08% to 44.43%; 2.39% to 15.16%; 3.58% to 7.04%; 2.34% to 3.80%; and 381.75 to 432.52 KJ/100g, respectively, as the supplementation of peanut and carrot flours increased. Conversely, the carbohydrate content decreased from 86.98% to 27.64%. The mineral content of the fortified samples also showed a significant increase (p<0.05) with higher levels of peanut and carrot flour supplementation. The findings of this study indicated a statistically reduction (p<0.05) in the antinutrient profiles of the fortified food samples. Additionally, the vitamin content significantly increased (p<0.05) with the addition of peanut and carrot flours. The overall viable counts were significantly low, and there were no detectable coliform or fungal counts. Although, the result of sensory evaluation indicated that the control sample was more organoleptically acceptable than the substituted samples, incorporating these nutrient-rich and functional ingredients in the production of maize-based cereals may broaden the application of peanut and carrot flours in the creation of various cereal types and other cereal-based food items.

A neuropharmacological profile of lycium schweinfurthii (solanaceae) methanolic extract in mice

Lycium schweinfurthii is a shrub belonging to the Solanaceae family which widely grows in North Africa and Mediterranean regions. The plant leaves have traditionally been used for gastrointestinal diseases as peptic ulcer in Libya. This study aimed to investigate the effect of Lycium schweinfurthii extract on the central nervous system in mice including anticonvulsant, antidepressant and muscle relaxant activities. The methanolic extract was prepared by fractionation technique. Albino male mice weighing 22 ± 2.0 gm were used and equally divided into equal number and weight for each experiment (n = 6). The best effective pharmacologically dose of 400 mg/kg, i.p. of the methanolic extract was selected to explore the anticonvulsant activity for picrotoxin-induced convulsion in mice (5.0 mg/kg), antidepressant activity of forced swimming test of depression and muscle relaxant action by motor coordination test of hanging wire. Fluoxetine (10.0 mg/kg), imipramine (15.0 mg/kg) and diazepam (5.0 mg/kg) were used as reference compounds. Lycium schweinfurthii extract exhibited a significant prolonged delay in the onset time of induced convulsion and significant decrease in the frequency of convulsion as well as a significant decrease in the duration time of attacks. Pretreatment with flumazenil (2.0 mg/kg) was found to increase the frequency and duration of convulsions without profound change in the onset time produced by Lycium schweinfurthii. For antidepressant activity, the plant leave extract significantly decreased immobility time duration without a muscle relaxant effect. The results suggest that the methanolic extract of Lycium schweinfurthii leave has anticonvulsant and antidepressant-like activities without any muscle relaxant effect in mice. Thus, Lycium schweinfurthii may have a neuropharmacological potential use in human.

Development of an optimal poly-1-order (op-1) model for approximating solar photovoltaic (pv) power generation

This study was conducted to develop and evaluate the Optimal Poly-1-Order (OP-1) model for approximating solar photovoltaic (PV) power generation. Using a mixed research method, the study employed Ibrahim’s simulation and prediction of grid-connected PV system theory with two objectives and their corresponding research questions. The study gathered primary and secondary data to approximate the implementation of a solar-PV system with an OP-1 model for generating electricity: optimizing energy production, load demands, and financial viability in the medical hostel facility of the University of Port Harcourt, Rivers State, Nigeria. With the use of simulation and descriptive methods of data analysis, results showed that the lighting system had 400 lights, each with 12W power. It operated for a total of 18 hours. Daily power consumption was 36,400 Wh. More so, it showed that 60 fans with 100W power were used during the same hours, resulting in a daily power usage of 108,000 Wh. Based on a comprehensive economic evaluation, the OP-1 solar-PV system was found to be economically viable for powering the medical hostel. The system met electricity demand, resulting in a remarkable 407% ROI and substantial savings for the grid, despite a lower optimized size of 193kW compared to the base peak generation of 383.90k. The study concluded and recommended that the proposed OP-1 Solar-PV power plant can meet the facility's electricity needs with a peak generation of 383.90kW and detailed energy analysis. Deploying this efficient solar-PV setup guarantees reliable and green electricity for the Medical Hostel, slashing the campus's carbon footprint and grid reliance.

Aqueous extract of hybanthus enneaspermus exhibited aphrodisiac potentials in fluoxetine-induced sexually-impaired female rats

Hybanthus enneaspermus, traditionally used as an aphrodisiac was investigated for its potential to reverse antidepressant-induced sexual dysfunction in female rats. The aqueous extract was evaluated for secondary metabolite, amino acid and mineral constituents. Alkaloids, tannins, flavonoids, anthraquinones, steroids, terpenoids, phenolics, calcium, potassium, sodium, glutamine and leucine are some of its notable constituents. 60 healthy, sexually responsive female albino rats (144.7±5.9 gm) were divided into six groups (A-F) of 10 rats each; of which 50 were induced into sexual dysfunction. Rats in group A were administered distilled water throughout the experimental period and served as a control group, while rats induced into sexual dysfunction (Groups B-F) by fluoxetine were given water, the reference medication (Tadalafil) and oral doses of the extract (250, 500, and 1000 mg/kg body weight) once daily for seven days, respectively. When administered to sexually active rats, fluoxetine significantly decreased the frequency of darting, hopping, lordosis, genital grooming, and licking behavior by 57.4%, 42.5%, 43.9%, 64.0%, and 41.8%, respectively. However, the latency of darting, hopping and lordosis were significantly increased by 50.6%, 47.7%, and 54.9%, respectively. Hybanthus enneaspermus aqueous extract administered at doses of 250, 500, and 1000 mg/kg significantly reversed fluoxetine-mediated changes in all sexual behavior parameters. The extract's ability to reverse the characteristics of sexual behavior at 1000 mg/kg was comparable to those of tadalafil-treated rats. Additionally, all the extract dosages reversed the levels of blood luteinizing hormone, follicle-stimulating hormone, progesterone, prolactin and estrogen after it has significantly been altered by fluoxetine. The results indicated that the aqueous extract of Hybanthus enneaspermus improved the proceptive, receptive and orientational behavior of rats. The extract also enhanced reproductive hormone concentration by restoring sexual competence in sexually-impaired female rats. The findings of this study provide further evidence in favor of Hybanthus enneaspermus widespread usage in the management of female sexual dysfunction.

Presentation and character for adult patients with diabetes in libya

Diabetes is a global issue, the diabetes epidemic is expected to continue, and the burden of diabetes causes catastrophic expenditure for healthcare system. The current study aimed to determine the presentation, the clinical feature and cardio-vascular risk factors in patients with diabetes. A retrospective observational study had been conducted in out-patients department at Almustaqpal Almosherq Centre during September, 2013 till September, 2020, the total number of attended out-patients department were 1 024, 820 patients who were selected for this study. A special perform was completed for every patient, which included details about patient’s demographics, points in clinical history, relevant investigations and clinical examinations were recorded. The study reported that out of 820 patients, 66% (n = 538) was female and their age range was between 14 - 87 years with a mean age of 56.53 ± 13.49 years, 96% (n = 791) were clinically diagnosed as type II diabetes, 07% of the patients were diagnosed as pre-diabetes, the duration of diabetes ranged from newly diagnosed to more than 10 years, with 46% (n = 379) of the studied population were more than 10 years diabetes duration, 70% (581) were presented with classical symptoms of diabetes. Initial treatment for diabetes also different in the studied sample, were absent of anti-diabetic medications in 30% (n = 248) of the patients, they refused to start glucose lowering drugs, 34.6% (n = 284) of them have morbid obesity (body mass index is more than 40), 80% (n = 662) have high HBA1c (more than 8 g%), 40.3% (n = 240/596) were uncontrolled hypertension on anti-hypertension drugs, 95.6% (n = 682/713) were controlled on treatment of lipid lowering drugs. This study showing the presentation of diabetes were the common, type II diabetes, at age group between 41 - 66 years about 65%, female sex, with high body mass index, high glycated hemglobulin and uncontrolled hypertension. There is concern that diabetic patients were occurring at a high frequency in younger adults, where longer duration of illness could increase the risk of developing more complications in later life. The rate of coexist cardiovascular risk factors (hypertension, dyslipidaemia and obesity) in Libyan patients with diabetes is highlighted.

Autophagy is a self-digesting mechanism responsible for removal of damaged organelles, malformed proteins during biosynthesis, and nonfunctional long-lived proteins by lysosome. Autophagy has been divided into three general types depending on the mechanism by which intracellular materials are delivered into lysosome for degradation that is, microautophagy, chaperone-mediated autophagy (CMA), and macroautophagy. In microautophagy cytoplasm material is sequestered through direct invagination to the lysosomal membrane. Whereas in CMA proteins flagged with pentapeptide motif (KFERQ) were selectively degraded through direct translocation into lysosome. Macroautophagy involves the formation of subcellular double-membrane-bound structures called autophagosomes that contain degradable contents of cytoplasm materials and deliver them into lysosomes for breakdown by lysosomal enzymes. The molecular mechanism of autophagy involves several conserved Atg (autophagy-related) proteins. Systems produce modified complexes Atg8-PE and Atg5-Atg12-Atg16 as autophagy regulators. Autophagy is activated in response to diverse stress and physiological conditions. For example, food deprivation, hyperthermia, and hypoxia are mediated by factors like insulin/IGF-1, m-TOR signaling, FOXO transcription factors, and chaperones. The perturbance in autophagy may lead to several types of cancers, myopathies, and neuromuscular disorders. Several autophagy inducers and inhibitors like 3-methyladenine (3-MA), bafilomycin A1, LY294002 (LY), and Velcade have been used to treat disease is an intense field of study.

The mechanistic insight of polyphenols in calcium oxalate urolithiasis mitigation

About 12% of the world's population is affected by different forms of urolithiasis, of which the recurrence rate in females is 47–60% and in males is 70–80%. Standard therapeutic agents (allopurinol, citrate, cystone and thiazide diuretics) prevent and treat urolithiasis. Still, these are not universally-effective due to common kidney stone relapse and other side effects. Surgical treatment causes long-term renal damage, hypertension and stone recurrence. Polyphenols, the plant-derived bioactive molecules, have showed protection against cancers, cardiovascular diseases, diabetes, osteoporosis and neurodegenerative diseases, among several other ailments. The role of these phytochemicals in urolithiasis management is emerging. Hence, the present review discusses peer-reviewed published literature to date on this aspect and highlights that polyphenols could effectively inhibit the formation of calcium oxalate urinary stones (most common renal stone), correlating with their antioxidant, anti-inflammatory, diuretic and angiotensin-converting enzyme (ACE) inhibition. Further, we have proposed the prospects and challenges in developing plant polyphenols into drugs against kidney stone prevention. This review might be a stepping stone for further investigation into the clinical implications of polyphenols in urolithiasis remediation.

The number of institutions and enrollment in higher education continue their rapid growth, but the quality of this education remains uncertain. A small number of state subsidized institutions attract a thin top layer of talent from each year’s cohort. High selectivity of admission to these elite institutions provides a screen valued by potential employers. Domestic and foreign demand for the services of these few thousand students has created an inflated reputation of the overall quality of India’s higher education. The number of such graduates remains small relative to the population and the demands of India’s economy for educated manpower. Reliable estimates of value-added by higher education, beyond the screening value of admission to elite institutions, are needed to assess colleges and universities, and to guide educational policy. Graduate education the seed farm of higher education and scholarship continues in an alarming state of disarray with respect to both quality and quantity. Pressed by budgetary constraints, the government appears to have decided on profit-oriented privatization of higher education as the solution. Political and business classes, with significant overlap between the two, see higher education as a source of lucrative private returns on investment. There is little theoretical or empirical evidence that supports the prospects of success of a for-profit model in building quality higher education. Some recent proposals hold promise of radical reform and renovation, including regulatory restructuring. It remains unclear whether the government has the wisdom, determination, financing, and power to push reforms past the resistance from entrenched faculty and from the political and business classes. It’s absolutely true that the future and overall development of any country depends upon the fact that how educated its citizens are. A review on the potential of remdesivir against sars cov2

Covid 19, the pandemic originated in the Chinese city of Wuhan, had the entire world conquered. The structure and transmission of the causative organism, Coronavirus is well studied. Remdesivir, the product of Gilead pharmaceuticals, was effective against many viral infections, including Ebola and SARS. It comes under the category of nucleoside prodrug and has given promising results in the early trials against SARS COV 19. In depth, research is taking place at a rapid pace, so that Remdesivir will be available to the therapeutic community as an effective remedy for the pandemic caused by SARS COV2. If this meets success, the darkest era in the modern history of mankind may become a memory in the near future.

“Phyllanthus emblica” known to be amla has role in the skin aging influences the changes in skin, including skin dryness, wrinkle, and irregular pigmentation. Initially the 6 day observation has been taken for the ageing activity to be track to study the TREM2 pathway of “Phyllanthus emblica”. Cellular observation and pathway consideration: The environmental impact of pH, Temperature, Humidity and stability of amla fruits is important for the ageing of cells in neuronal cascade of TREM2 Pathway, while studying the fruits cell cycle. The melanin suppression through inhibition of tyrosinase and tyrosinase-related protein-2 activities, the strong antioxidant, and the potent matrix metalloproteinase-2 in cellular observation of tyrosinase pathway. The study aimed to evaluate the anti-skin aging efficacy of amla.

Urolithiasis is a common worldwide problem with high recurrence. This review covers forty four (44) families starting from alphabet A to L and includes Bignoniaceae (05); Araceae, Burseraceae, Combretaceae (04); Annonaceae, Berberidaceae, Betulaceae, Gentianaceae, Gesneriaceae (03); Aizoaceae, Adiantaceae, Alismataceae, Aristolochiaceae, Asclepiadaceae, Bombacaceae, Cannabaceae, Cyperaceae, Geraniaceae and Iridaceae (02); Acoraceae, Adoxaceae, Armatellaceae, Aquifoliaceae, Araliaceae, Averrhoeaceae, Basellaceae, Begoniaceae, Bromeliaceae, Cactaceae, Calophyllaceae, Campanulaceae, Caprifoliaceae, Caricaceae, Celastraceae, Clusiaceae, Dracaenaceae, Dryopteridaceae, Elaeagnaceae, Grossulariaceae, Hyacinthaceae, Hydrangeaceae, Hypoxidaceae, llecebraceae and Juglandaceae (01) plant used globally in different countries. The plants of three families Asteraceae, Fabaceae and Lamiaceae are revisited to provide updated information. This review will not only be useful for the general public but also attract the scientific world for antiurolithiatic drug discovery.
